Correlation of Histopathologic/Cytologic Follow-up Findings With Vaginal ASC-US and ASC-H Papanicolaou Test and HPV Test Results.

Abstract
Current American Society of Colposcopy and Cervical Pathology recommendations about human papillomavirus (HPV) triage and further management for atypical squamous cells are pertinent to cervical Papanicolaou (Pap) tests. There are limited data on HPV detection in vaginal liquid-based cytology (LBC) specimens. The aims of this study were to determine whether adjunctive high-risk (HR)-HPV testing is useful for disease risk assessment in women with vaginal atypical squamous cells of undetermined significance (ASC-US) and atypical squamous cells, cannot exclude HSIL (ASC-H) Pap results. We identified 1,125 ASC-US and 36 ASC-H vaginal Pap results with HR-HPV testing. Of the cases, 244 (21.7%) ASC-US and 21 (58%) ASC-H were HR-HPV+. Among ASC-US H...
Cytomorphologic diagnosis and HPV testing of metastatic and primary oropharyngeal squamous cell carcinomas: A review and summary of the literature
Mon, 20 Feb 2012 05:00:00 +0100 | Diagnostic Cytopathology
AbstractA significant proportion of squamous cell carcinomas (SCC) of the oropharynx are associated with high‐risk human papillomavirus (HPV). These HPV‐related carcinomas are prognostically and therapeutically different from traditional SCC of the head and neck. Cervical metastases from head and neck primary SCC are frequently investigated by fine‐needle aspiration (FNA). HPV testing of cytologic samples from these lesions would potentially simplify diagnosis and therapeutic planning. This review summarizes current methodologies for HPV testing of FNA specimens and the role of HPV typing in diagnosis, prediction of prognosis, and selection of therapy. Diagn. Human papillomavirus detection in dysplastic and malignant oral verrucous lesions
Fri, 17 Feb 2012 05:00:00 +0100 | Journal of Clinical Pathology
This study used a multi-tiered method of HPV detection using DNA in-situ hybridisation (ISH) for low- and high-risk subtypes, consensus PCR, and HPV genotype analysis in archival tissue from 20 cases of dysplastic and malignant oral verrucous lesions. The biological significance of HPV DNA detection was assessed by p16 immunohistochemistry (IHC). While 1/7 carcinomas and 5/13 dysplasias contained HPV DNA by consensus PCR and genotype analysis, all specimens were negative for low- and high-risk HPV ISH and negative for p16 IHC. Results show that although high-risk HPV DNA is detectable in a subset of these lesions, the lack of p16 overexpression suggests that the oncogenic process is not driven by HPV oncoproteins. A point mutation in the DNA-binding domain of HPV-2 E2 protein increases its DNA-binding capacity and reverses its transcriptional regulatory activity on the viral early promoter

Conclusions: These results suggest that the mutation from Ala to Val at aa 338 is critical for E2 DNA-binding and its transcriptional regulation. (Source: BMC Molecular Biology - Latest articles)
Cervical cytological changes in HIV-infected patients attending care and treatment clinic at Muhimbili National Hospital, Dar es Salaam, Tanzania
Wed, 15 Feb 2012 05:00:00 +0100 | Infectious Agents and Cancer
Background:
Tanzania is among Sub-Saharan countries mostly affected by the HIV and ---- pandemic, females being more vulnerable than males. HIV infected women appear to have a higher rate of persistent infection by high risk types of human papillomavirus (HPV) strongly associated with high-grade squamous intraepithelial lesions (HSIL) and invasive cervical carcinoma. Furthermore, although HIV infection and cervical cancer are major public health problems, the frequency and HIV/HPV association of cervical cancer and HSIL is not well documented in Tanzania, thus limiting the development of preventive and therapeutic strategies.
Methods:
A prospective unmatched, case-control study of HIV-seropositive, [greater than or equal to]18 years of age and consenting non-pregnant patients attending the...
